Bacteriostatic vs. sterile water: which to use

When reconstituting a lyophilized peptide, the liquid you choose changes how long you can use that vial. The two usual options — bacteriostatic water and sterile water — look interchangeable, but they behave differently. The difference comes down to a preservative.

Sterile water

Sterile water is purified water from which microorganisms have been removed, with nothing added. It's clean the moment you open it, but it has no way to stay clean afterward. As soon as you pierce the stopper and insert a needle, contamination becomes possible, and the water does nothing to stop it. It works well when you're going to use the entire contents at once.

Bacteriostatic water

Bacteriostatic water is sterile water with a preservative, usually 0.9% benzyl alcohol. That preservative inhibits bacterial growth, so the vial tolerates several punctures over time without a minor slip ruining it. It's the standard choice when a reconstituted vial will be used across several draws over days or weeks.

For most multi-use reconstitution work, bacteriostatic water is the practical choice. Sterile water makes sense when the protocol consumes the entire vial in a single session.

How it affects your calculation

The type of water doesn't change the arithmetic of the concentration — that depends on the milligrams of peptide and the volume you add — but it does change the volume worth using. If you're going to draw from the vial many times, a larger volume of bacteriostatic water makes it easier to measure small doses precisely in the syringe. If the vial is single-use, you can reconstitute in less liquid.
